Advancing STED super-resolution optical microscopy with differential depletion

In this presentation, we introduce two simple and easy-to-implement methods that can efficiently suppress the low spatial frequency background appearing in STED images. Polarization switching STED subtracts the background noise by obtaining background-only images by using switched polarization light beam. Differential stimulated emission depletion decomposes the background noise by altering the STED illumination intensities, and it achieves high signal-to-background ratio and spatial resolution at the same time.
